The average of the threshold cycles was used to interpolate standard curves and to calculate the transcript
amount in samples using SDS software (v.2.2) (Applied Biosystems). CD8+ T cells (≥98% pure) were obtained by positive magnetic selection from pooled spleens as above. For flow cytometry experiments, cells (1 × 105 cells/well) were cultured at 37°C, 5% CO2 in 96-well, flat-bottomed plates (BD Labware, Badford, MA, USA) in 200 μL of RPMI 1640 medium (Cambrex, Baltimore, MD, check details USA) containing L-glutamine supplemented with 10% FCS, antibiotics, β-mercaptoethanol (Medium). Cells were incubated with medium alone or with recombinant mouse IL-15, IL-7, and TSLP (all from R&D). For real-time PCR experiments, cells were cultured as above, except that they were incubated in 24-well plates (5 × 106 cells/well). Statistical analysis was performed by a Student’s t-test. “Human papillomavirus (HPV) infections account for more than 50% of infection-linked cancers in women worldwide. The immune system controls, at least partially, viral infection and around 90% of HPV-infected women clear the virus within two years. However, it remains unclear which immune cells are implicated in this process and no study has evaluated the direct interaction between HPVs and NK cells, a key player in host resistance to viruses and tumors. We demonstrated an NK-cell infiltration in HPV-associated preneoplastic cervical lesions.
